“Mass Defect” Tags for Biomolecular Mass Spectrometry

Presented at BIO 2005 Annual International Convention, June 19-22, 2005, Philadelphia Pennsylvania

Abstract
Luke V. Schneider, Michael P. Hall, Lane A. Clizbe, John P. Wilson, Siamak Ashrafi, Robert Petesch, Christine Hobson, and David Bomberger

The mass defect effectively "shifts" the peaks of tagged biomolecules relative to untagged biomolecules in the mass spectrogram by –0.1 amu, allowing their discrimination. Mass defect tagging technology has may applications, such as:
  • Low cost and quantitative protein and DNA microarray experiments with the advantage of mass spectrometric identification, quantitation, and characterization of the bound species.
  • Protein and glycan sequencing at 10-100 fold speed and cost advantages over current methods, with significantly less ambiguous results.
  • Top-down characterization of unknown proteins.
